CONVENTIONAL FLOW CYTOMETRY
BD LSR Fortessa (6-laser)
  • 6-laser cell analyser

  • Lasers: 355 nm, 405 nm, 488 nm, 545nm, 561 nm, 628 mn. One of the lasers is a B/V 545nm laser for detection of AmCyan

  • 20 parameters: 18 colours, 2 scatter

  • Speed: sample acquisition of 10,000 events/sec

  • Capabilities: Samples from 5 mL tubes

  • 5-laser SPECTRAL analyser

  • Capabilities: Spectral unmixing of highly-overlapping fluorophores, autofluorescence subtraction. Three instruments include plate sample loaders with mixing and temperature control

CELL SORTING

 

BD InFlux (6-laser)
  • Stream in air sorter

  • Lasers: 355nm, 405nm, 445nm, 488nm, 552nm and 640nm

  • 21 colour + 3 scatter, including the Small Particle Detector (2 FSC PMTs)

  • Capabilities: 

    • 24 parameters (up to 22 colours, 2-3 scatter, including polarised FSC)

    • Up to 30,000 events/sec

    • Supports 2-, 4- and 6-way cell sorting 

    • Fully enclosed in a certified Class II hood and sort sterility is a high priority. Cooled sample delivery and collection

    • 1.7mL, 5mL, 15mL and 50mL tube holders for sort samples, plates (6-384well), slides

    • Index sorting

    • Computerised Cell Deposition Unit (CCDU) allowing for single-cell sorting into 384 well-, 96 well- and PCR-plates, or onto microscope slides

Full Spectrum Cell Sorter: Cytek Aurora CS
  • Stream in air spectral sorter

  • Lasers: 351 nm (16 detectors), 405 nm (16 detectors), 488 nm (14 detectors), 561 nm (10 detectors), 638 nm (8 detectors). Three scattering channels and 64 fluorescence channels.

  • Capabilities: 

    • Sort up to 6 ways into 1.5 mL tubes, or up to 4 ways into 5 mL tubes. 

    • 40 colours demonstrated including fluorochromes with emission spectra in close proximity to each other.  

    • Customizable nozzle settings and sort modes

    • Automated drop delay, sort monitoring, and clog detection for a reliable sorting experience.

    • Extract autofluorescence and improve resolution of highly autofluorescent samples.

    • Choose from a variety of sample input and collection devices including 5 and 15 mL tubes for input and 96-well plates, 1.5 and 5 mL tubes for collection. Sort into 96-well plates.

IMMUNOASSAYS (LUMINEX)

The Luminex is an instrument that enables multiplexing of different analytes in a single microplate well for biological assays similar to ELISA. It is based on x-MAP microsphere technology, where beads are fluorescently coded such that beads reactive to different analytes have known, distinct fluorescent properties allowing them to be analysed simultaneously.

 

Analytes of interest are sandwiched between these coded beads and a reporter antibody, allowing quantification of analyte concentration. The Luminex 200 system housed at MIMR can analyse up to 80 analytes simultaneously, with minimal volume required (10-20uL) and a plate read time of approximately 45 minutes.
